Abstract

Cassava waste consists of two parts : the cassava skin which is known as brown skin, and the white part which is attached to the brown skin. We are trying to utilize the cassava waste for making glucose as a material for bioethanol. The aim of this experiment is to learn the effect of slurry concentration and calcium ions concentration to cassava waste hydrolysis. There are two steps in this hydrolysis : liquefaction process using amylase and saccharification process using glucoamylase. The liquefaction process was conducted at 95oC and the saccharification process was conducted at 60oC using Erlenmeyer flask as container for the media and cassava waste. The glucose concentration has been analyzed using DNS method. There was also qualitative analysis of calcium ions to determine how much calcium ions are utilized by enzyme. The experiment result shows that the higher concentration of slurry and calcium ions introduced to the media, the higher glucose concentration will be obtained. The optimum condition for this hydrolysis process cannot be determined yet, since the profile of glucose concentration, CaCl2 concentration and slurry concentration is continuously increased at the range data taken in the experiment which is shown by surface response method. Hence, broader range of the data will be needed for the next experiment to find the optimum condition. It is shown that the higher concentration of calcium will result in the higher enzyme activity. Highest concentration of glucose is obtained from 32% w/v slurry concentration and 50 ppm CaCl2 concentration which is 238,701 gram/L. All Calcium ion was used by enzyme to maintain its activity.
