Abstract
The ethanolic extract of Moringa oleifera leaf exhibited high antioxidant activity, nevertheless the distinctive odor, bitter taste, hygroscopic character, and limited solubility in water are the limitations of preparing this extract into pharmaceutical products. Solid dispersion is an attempt for improving the physical characteristics of these extract and enhance the solubility in water. This study aims to develop solid dispersion containing Moringa oleifera leaf extract using non-solvent method called microwave irradiation. Hydrophilic carrier such as mannitol was employed in this study to increase the wettability and improve the physical characteristics of extract. The power and time exposure of microwave energy exhibited pivotal role in the crystalline phase conversion into amorphous state, consequently influenced the solubility and dissolution rate of Moringa oleifera leaf extract. In this study, solid dispersion of Moringa oleifera leaf extract-mannitol in 1:4 ratio has been prepared by different combination of power (300 W and 400 W) and exposure time (2 minutes and 3 minutes). Physical and mechanical characteristics of solid dispersion powder containing the extract exhibited good flowability and compressibility. The moisture content was ranged between 2.23-2.45%. The results of thermal characterization by differential scanning calorimetry (DSC) showed a decrease of crystal lattice energy of the solid dispersion. Crystallinity analysis using powder X-ray diffraction (PXRD) exhibited intensity reduction of distinctive peaks represent decrease in crystal lattice regularity. The results of scanning electron micrograph indicated that the extract entrapped in the mannitol structure in the form of equant-shaped particles and agglomerates, meanwhile the physical mixture was captured as aggregates and agglomerates particle. The hydrogen bonding between the extract and mannitol has been confirmed from the fourier-transform infrared spectrum. Preparation of Moringa leaf extract solid dispersion by 400 W and 3 minutes exposure time was the optimized condition due to lowest moisture content, better flowability, and the significant reduction of crystal lattice regularity.
