Abstract
Background: Toll-like receptors (TLRs) are generally expressed in the central nervous system (CNS) and play an important role in cell survival, including neurons, microglia, astrocytes, and oligodendrocytes. TLR2 is one of the most studied TLR groups and is associated with neurodegenerative diseases because it can cause pro- inflammatory conditions in the CNS. Phytoestrogen compounds are able to replace the role of estrogen in maintaining body homeostasis, including in the CNS. This study aims to determine the role of phytoestrogen compounds in inhibiting TLR2 (6NIG) activation, which can cause neurodegenerative diseases. Method: The method used is molecular docking with the AutoDockTools 1.5.6 program. Predictions of pharmacokinetic and pharmacodynamic properties were carried out using SwissADME, while for toxicity using pkCSM and ProTox II. Results and Discussion: The docking results using the 6NIG protein revealed that the compounds α-amyrin and β-amyrin had the highest binding potential when compared to the positive control, 17β-estradiol. Pharmacokinetic and pharmacodynamic tests showed that both compounds have good permeability, strong lipophilicity so they can penetrate cell membranes, and are not toxic. Conclusion: the compound has the potential to treat neurodegenerative diseases by targeting the TLR2 (6NIG) receptor.
