Abstract
Research was conducted in order to detect active chemical constituent of Typhonium flagelliforme which has antioxidant property and at once, also able to interact with DNA. Active extract was determined by evaluation of antioxidant activity using DPPH. The most active extract of Typhonium flagelliforme was further purified by column chromatography. Active fraction is partially analyzed and visualized at 365 nm and sprayed by visualization reagent to detect alkaloids, terpenoids and flavonoids qualitatively. The capability of interaction with DNA was investigated by thermal denaturation assay. The result showed that highest antioxidant activity is given by dichloromethane extract with an IC50 = 245.989 ppm. The most active fraction was obtained from the root with an IC50=231.199 ppm. Thermal denaturation assay showed interaction between DNA and constituent in most active fraction. Alteration in DNA melting temperature (Tm) indicated interaction between DNA and the constituents. More than one active constituent was indicated by TLC analysis. Phytochemistry analysis showed that the most active fraction contains alkaloids, terpenoids and flavonoids.
