Abstract
Klebsiella pneumoniae is one of pathogenic bacteria known to be able to detoxify cadmium. Cadmium -resistant Klebsiella pneumoniae is usually isolated from-high cadmium areas. In this study, Klebsiella pneumoniae was isolated from human oral cavity to study cadmium detoxification by cysteine desulfhydrase. The aim of this research is to clone cysteine desulfhydrase gene of Klebsiella pneumoniae isolated from oral cavity into Escherichia coli. The result showed that primer was designed and succesfully been used to amplify cysteine desulfhydrase gene from K. pneumoniae genome. The gene was cloned into pUCP-18. The recombinant plasmid was successfully transformed into Escherichia coli DH5α using the heat shock transformation method and screened on LB-agar-IPTG-Cd plate. There were eight transformant colonies were able to grow on Luria-Bertani agar containing 75 ug.mL-1 cadmium such as the K. pneumoniae original cells. E. coli wild type was not able to grow at those concentration of cadmium. As conclusion that the cloning of cysteine desulfhydrase gene from K. pneumoniae isolated from oral human was succesfully been done and gave cadmium resistance to E. coli cell
